Peptide Predictions

Peptides represent Protein - Peptide Antibodies Our scientists work with you to identify peptides that represent your protein in accordance with Davids peptide prediction algorithms (see info box). Use this free service by sending us an email with your one-letter amino acid sequence (txt, pdf, fasta or directly in the email). All of your information is kept private. We will only begin synthesis of your peptide after you have confirmed one or two of the proposed peptides for antibody generation.
In collaboration with our experts, our prediction algorithm suggests peptides that represent your protein and are ideal for antibody generation. These peptides must be antigenic, soluble, and have high epitope surface area. You can immunize one animal with two distinct peptides to increase your chances of using the peptide that best represents your protein. The likelihood of using the optimal peptide rises from 70% (1 peptide) to 90% (2 peptides) to 97%. (3 peptides).


Peptide Synthesis

Solid phase synthesis is used to produce peptides. Each peptide passes quality control using HPLC and MS. For immunizations, the purity ranges from 70 to 98 percent. If you want a higher purity, please request this service. The services described in the table below are designed specifically for antibody generation. Davids "All-Inclusive-Service" includes everything you need, including peptide synthesis and conjugation. Please request a different amount and purity of a peptide or just a synthesis, if you don't need antibodies.

Peptide Conjugation

Peptides are covalently conjugated to a carrier such as Key Limphole Hemocyanin (KLH), Ovalbumin, Thyroglobulin, Bovine Serum Albumin (BSA), or other molecules to increase antigen presentation from small molecules. Following immunizations, affinity purification is recommended in order to get monospecific antibodies. The conjugation is performed at the -NH2, -COOH, -SH, -OH or other groups of the amino acids, depending which amino acids are available at the peptide. To enhance the antigenicity multiple carriers or multiple conjugation methods can be used. This Mixed Conjugation (P111) includes the peptide synthesis and is recommended for peptides with low antigenicity. Davids All-Inclusive-Services P101 and P102 include the peptide synthesis, conjugation and quality control as well.

Peptide Immunization

Davids begins vaccinations as soon as the peptide synthesis and preparation is completed. Please include one of our custom polyclonal antibody services for rabbits, chicken, mice, rats, guinea pigs, or one of our custom monoclonal antibody services for Mice or Rats in your purchase.
To get monospecific antibodies following peptide vaccination, we recommend affinity purification for polyclonal antibodies.

Additional Services

When using peptides for in vivo cell culture applications, additional services such as peptide preparation are recommended. The peptide fraction is diluted here, and impurities are removed using HPLC. Lyophilization is used to eliminate vaporous compounds. In a third stage, the peptide fraction is sterile filtered and diluted in a cell culture suitable buffer such as PBS or water.
